Human IL5 is measured using sandwich immunoassay involving two monoclonal antibodies, one labelled with Eu-Cryptate (Donor) and the second with d2 (Acceptor). The intensity of the signal is proportional to the concentration of the IL5 present in the sample.

A Human IL5 test, like all cytokine assays, can be run with cell supernatants or with whole cells using the protocol described on the left. The antibodies labelled with HTRF? fluorophores may be pre-mixed and added in a single dispensing step to further streamline the protocol. An assay can be run in 96- to 384-well plates by simply resizing each addition volume proportionally. The Human IL5 assay protocol is described here, using a white 384-well small volume plate.

As described on the figure on the right, the performances of the assay are not affected by culture media (RPMI + 10% FBS)
